Protocol for Parental Cell Line (GM25256)

The protocol you provide for your cell lines (GM25256*l) is slightly different from the protocol provided for the parental line (GM2526). My lab is interested in purchasing and culturing both your cell lines and the parental, but we prefer your protocol since it uses pen/strap. Did/do you use the same protocol you have developed when you cultured the parental line?

Hi Lauren, you can add Pen Strep to your cultures (that would not make a difference). We recommend thawing and culturing our cell lines in mTESR. You are welcome to adapt the cultures using different media (if your lab uses something else). It is also highly recommended to freeze back working banks of each cell line you plan to purchase prior to making any changes to media or matrices. If you have purchased the parental line from Coriell please be sure to follow their thaw instructions (thaw in 1 well of 6 well Matrigel coated plate). However, our vials from our Allen Cell Collection will need to be thawed into 1x10 cm Matrigel coated plate.

Also, our protocol is a single cell passage method. The cells in the Allen Cell Collection were cultured and frozen as single cells. I believe Coriell cultures in clump passaging and why they recommend versene rather than accutase.
