How do you to keep cardiac differentiated cells from coming apart and lifting from well? My cells beat so hard they do not last that long, a month a most.
Hi, thanks for your question. We typically replate the cardiomyocytes on Day 12 of the differentiation protocol to avoid this issue. For our purposes, we typically replate onto mouse laminin-coated glass bottom plates, but other researchers have replated onto plastic as well to keep the cells in culture for other purposes. You can find the details of our replate protocol in the Methods section of this 2021 Cell Reports paper: https://www.cell.com/cell-systems/fulltext/S2405-4712(21)00156-3
