After you edit the cells, how do you test for pluripotency of the hiPSCs?
We look for expression of pluripotency markers like Oct3/4, Sox2, and Nanog by flow cytometry. We also differentiate iPSCs into the three germ layers using a commercial kit and look for markers of ectoderm, endoderm and mesoderm by droplet digital PCR or flow cytometry.
