Your RNP Transfection in WTCs protocol lists the use of the Neon 100uL kit, but I was hoping to use the 10uL kit. In your experience with the dCas9-KRAB cells, is the transfection efficiency high enough that the smaller volume kit can be utilized? Are there any other reasons why the 100uL kit should be used?
Thanks for the question. We do not have an answer as we have not tried it. However, we have tried the Lonza nucleofector device and that is successful, and is smaller volume. We would expect the Neon 10uL to work just as well. From a formal point of view, we do not have experience to report.
